Heat identification by 17&#946;-estradiol and progesterone quantification in individual raw milk samples by enzyme immunoassay Electron. J. Biotechnol.
Domènech,Anna; Pich,Sara; Arís,Anna; Plasencia,Carmen; Bach,Alex; Serrano,Alicia.
Background: There is a substantial decline in first-service-pregnancy-rate in dairy cows. In this regard, future prospects are to measure milk hormones on-farm and progesterone levels in milk are not enough to precise ovulation unless connected to other data. The objectives of this study were to investigate whether 17&#946;-estradiol could be measured from individual cow milk samples using a commercially available non-radiolabelled enzyme immunoassay kit (EIA) with no previously reported milk application, and whether those detections could precisely illustrate 17&#946;-estradiol pre-ovulation peak in spite of its limited concentration and short manifestation in milk. Results: Milk sample treatments for progesterone and 17&#946;-estradiol EIA...

ECONOMIC AND FINANCIAL VIABILITY OF DIGESTER USE IN CATTLE CONFINEMENT FOR BEEF REA
Montoro,Stela B.; Santos,David F. L.; Lucas Junior,Jorge de.
ABSTRACT This research analyzes the economic and financial feasibility of implementing a biodigester for confinement of beef cattle on a farm located in the State of São Paulo, through the approach of discounted cash flow. The anaerobic digestion technology arouses interest, due to its advantages with the generation of electricity and the pollution reduction of water resources, soil and air, from the biogas and bio-fertilizer production, high value-added products to cattle raising. It was verified that the treatment system of cattle manure with biodigesters generate biogas and fertilizer as revenue, with free cash flows that enable a return on investment in 3.8 years and return an IRR 26.40% and an MIRR of 10.69%; all above than the minimum rate of...

The effect of the bovine major histocompatibility complex (BoLA) locus on animal health may be due to a direct action of its alleles on immune functions, whereas its indirect effect on production traits might be explained by the better general health conditions of more productive animals. In the present study, the BoLA-DRB3 gene was investigated in 1058 cows belonging to seven Brazilian Gyr Dairy herds (Bos indicus, Zebu cattle). A total of 37 alleles were identified, 15 of them described for the first time in a Zebu breed. A highly significant association (p < 0.02) was observed between allele *54 and a decrease (-26.1 kg) in milk protein yield and there was a significant association (p < 0.05) between this allele and lower (-26.07 kg) milk fat...

Phylogenetic position of an uncharacterized Brazilian strain of bovine papillomavirus in the genus Xipapillomavirus based on sequencing of the L1 open reading frame Genet. Mol. Biol.
Lunardi,Michele; Claus,Marlise P.; Alfieri,Amauri A.; Fungaro,Maria Helena P.; Alfieri,Alice F..
The use of PCR assays with degenerate primers has suggested the existence of numerous as yet uncharacterized bovine papillomaviruses (BPV). Despite the endemic nature of BPV infections, the identification of BPV types in Brazilian cattle is still only sporadic. However, in a recent analysis of a partial segment of the L1 gene, we observed notable diversity among the BPV types detected. The aim of this study was to determine the phylogenetic position of the previously identified wild strain BPV/BR-UEL2 detected in the state of Paraná in Brazil. Since previous analysis of the partial L1 sequence had shown that this strain was most closely related to BPV type 4, genus-specific primers were designed. Phylogenetic analysis using complete L1 ORF sequences...

Zero-inflated Poisson regression models for QTL mapping applied to tick-resistance in a Gyr x Holstein F2 population Genet. Mol. Biol.
Silva,Fabyano Fonseca; Tunin,Karen P.; Rosa,Guilherme J.M.; Silva,Marcos V.B. da; Azevedo,Ana Luisa Souza; Verneque,Rui da Silva; Machado,Marco Antonio; Packer,Irineu Umberto.
Nowadays, an important and interesting alternative in the control of tick-infestation in cattle is to select resistant animals, and identify the respective quantitative trait loci (QTLs) and DNA markers, for posterior use in breeding programs. The number of ticks/animal is characterized as a discrete-counting trait, which could potentially follow Poisson distribution. However, in the case of an excess of zeros, due to the occurrence of several noninfected animals, zero-inflated Poisson and generalized zero-inflated distribution (GZIP) may provide a better description of the data. Thus, the objective here was to compare through simulation, Poisson and ZIP models (simple and generalized) with classical approaches, for QTL mapping with counting phenotypes...

Quantification of bovine cytokine gene expression using real-time RT-PCR methodology Genet. Mol. Biol.
Zaros,Lilian Giotto; Bricarello,Patrízia Ana; Amarante,Alessandro Francisco Talamini; Coutinho,Luiz Lehmann.
T cells produce cytokines that affect host response to infection. This paper reports real-time RT-PCR conditions and validation steps for accurate quantification of Bos indicus cytokines, interleukin (IL)-2, IL-4, IL-8, IL12p-35, IL-13, tumoral necrosis factor (TNF)-alpha, interferon (IFN)-gamma, monocyte chemoattractant proteins (MCP)-1 and MCP-2, and the glycoprotein mucin (MUC)-1 in two groups of Nelore cattle, one resistant and the other susceptible to gastrointestinal nematode infections. RPL-19 was shown to be an ideal internal control gene, since its expression was constant across treatments and presented lower variation when compared to the GAPDH gene. The optimized conditions established in the present study can be used to determine the immune...

Implementation of a parentage control system in Portuguese beef-cattle with a panel of microsatellite markers Genet. Mol. Biol.
Carolino,Inês; Sousa,Conceição O.; Ferreira,Sónia; Carolino,Nuno; Silva,Fátima S.; Gama,Luís T..
A study was conducted to assess the feasibility of applying a panel of 10 microsatellite markers in parentage control of beef cattle in Portugal. In the first stage, DNA samples were collected from 475 randomly selected animals of the Charolais, Limousin and Preta breeds. Across breeds and genetic markers, means for average number of alleles, effective number of alleles, expected heterozygosity and polymorphic information content, were 8.20, 4.43, 0.733 and 0.70, respectively. Enlightenment from the various markers differed among breeds, but the set of 10 markers resulted in a combined probability above 0.9995 in the ability to exclude a random putative parent. The marker-set thus developed was later used for parentage control in a group of 140 calves from...

Use of the TSPY gene for sexing cattle Genet. Mol. Biol.
Lemos,Daniela Cristina; Rios,Álvaro Fabrício Lopes; Caetano,Lisandra Cristina; Lôbo,Raysildo Barbosa; Vila,Reginaldo Aparecido; Martelli,Lúcia; Takeuchi,Paula Lumy; Ramos,Ester Silveira.
The Y-encoded, testis-specific protein (TSPY) is a Y-specific gene. The copy numbers of TSPY range from 20 to 60 in men and up to 200 in bulls. In this study, we examined the possibility of using the TSPY gene to sex cattle. DNA from blood samples of 100 Nelore cattle (50 males and 50 females) from the Nelore Cattle Breeding Program (PMGRN) was screened for TSPY by PCR using TSPY-specific primers. The assay was highly specific since all male samples were TSPY-positive and all female samples were negative. Positive results were also obtained at low DNA concentrations (less than 1 rhog/muL). These results showed that TSPY was a good male-specific marker, the usefulness of which was enhanced by the high copy number of the gene. This is the first report to...

Quantitative trait locus affecting birth weight on bovine chromosome 5 in a F2 Gyr x Holstein population Genet. Mol. Biol.
Gasparin,Gustavo; Miyata,Marcelo; Coutinho,Luiz Lehmann; Martinez,Mário Luiz; Silva,Marcos Vinícius G. Barbosa da; Machado,Marco Antônio; Campos,Ana Lúcia; Regitano,Luciana Correia de Almeida.
Segregation between a genetic marker and a locus influencing a quantitative trait in a well delineated population is the basis for success in mapping quantitative trait loci (QTL). To detect bovine chromosome 5 (BTA5) birth weight QTL we genotyped 294 F2 Gyr (Bos indicus) x Holstein (Bos taurus) crossbreed cattle for five microsatellite markers. A linkage map was constructed for the markers and an interval analysis for the presence of QTL was performed. The linkage map indicated differences in the order of two markers relative to the reference map (<A HREF="http://www.marc.usda.gov/">http://www.marc.usda.gov</A>). Interval analysis detected a QTL controlling birth weight (p < 0.01) at 69 centimorgans (cM) from the most centromeric marker...

Analysis of tRNA halves (tsRNAs) in serum from cattle challenged with bovine viral diarrhea virus Genet. Mol. Biol.
Taxis,Tasia M.; Bauermann,Fernando V.; Ridpath,Julia F.; Casas,Eduardo.
Abstract Acute infections of bovine viral diarrhea virus (BVDV) lead to a range of clinical presentations. Laboratory tests for detection depend on collection of samples during a short viremia. Acutely infected animals remain largely undiagnosed. Transfer RNA halves (tsRNAs) are hypothesized to function like microRNAs to regulate gene expression during an immune response. The objective of this study was to identify tsRNAs in cattle that had been challenged with a non-cytopathic field strain of BVDV. Colostrum-deprived neonatal Holstein calves were either challenged with BVDV (n=5) or mock challenged (n=4). Sera was collected prior to challenge and days 4, 9, and 16 post challenge. RNA was extracted and read counts of small non-coding RNAs were assessed...
